چکیده

A method for quantitative analysis of free amino acids in rat brain was established. Rat midbrain was homogenized in HC104 solution and the supernatant was applied to a high-performance liquid chromatograph attached to a fluorescence detector. Amino acids were separated in an ion exchange column prepared for amino acid analysis and were reacted with o-phthalaldehyde. Fluorescence intensity was linear in a wide range of amounts of authentic amino acids (1.25-12.5 nmoles). Threonine, serine and glutamine were completely separated. Sixty minutes was required for the elution of nine amino acids, which were main components of free amino acids in rat midbrain. An 8-hr exposure of rats to 4000 ppm of toluene or hexane produced significant increase in glutamine content of the midbrain.

اثرات مختلف استرس حاد و مزمن ناشی از بی‌حرکتی روی آزمون فرمالین در موش صحرایی نر

Background and Objectives : Acute and chronic stress induces hormonal and neuronal changes which affect both pain threshold and nociceptive behaviors. But the effect of acute and chronic immobilization stress on formalin induced nociceptive behaviors are unknown. Therefore, this study evaluated the effects of acute and chronic immobilization stress formalin test on the male rat.   Material an...

Association of morphine-induced analgesic tolerance with changes in gene expression of GluN1 and MOR1 in rat spinal cord and midbrain

Objective(s): We aimed to examine association of gene expression of MOR1 and GluN1 at mRNA level in the lumbosacral cord and midbrain with morphine tolerance in male Wistar rats. Materials and Methods: Analgesic effects of morphine administrated intraperitoneally at doses of 0.1, 1, 5 and 10 mg/kg were examined using a hot plate test in rats with and without a history of 15 days morphine (10 mg...

Chronic exposure to toluene changes the sleep-wake pattern and brain monoamine content in rats.

Toluene, found in glues and cleaners, is among the inhalants most commonly abused by workers and young drug addicts. In this study, we examined the changes in sleep patterns and monoamine content induced by chronic toluene exposure. Rats were chronically exposed to toluene vapors beginning at 30 days of age for a duration of 30 days. Experiment I was performed in a control group (n=10) and a ch...

Changes in Catecholamines and Acetylcholinesterase Levels of Crebellum, Mid-brain and Brain Cortex in Chromium Treated Rats

The short and long term effects of chromium toxicity on brain catecholamines and acetylcholinesterase levels were investigated. Rats were injected daily with varying amounts of chromium. The short term (2 h) administration of chromium (8 mmol/kg) reduced catecholamines level of cerebellum, mid-brain and brain-cortex by 22.8, 19.4 and 21.2% respectively. Acetylcholinesterase activity was also re...

Morphine-induced analgesic tolerance is associated with alteration of protein kinase Cγ and transient receptor potential vanilloid type 1 genes expression in rat lumbosacral cord and midbrain

Introduction: Transient receptor potential vanilloid type 1 (TRPV1) and protein kinase Cγ (PKCγ) are involved in sensitization/desensitization to noxious stimuli. We aimed to examine the gene expression levels of TRPV1 and PKCγ in rat lumbosacral cord and midbrain on days 1, 4 and 8 of induction of morphine analgesic tolerance. Methods: Two groups of male Wistar rats received ...
